Detection of Translocatio Using Dual Fluorescence Probe on Interphase Nuclei


Abstract
Background :
@EN Conventional banding techniques are sometimes very difficult to get the good methaphase from cultured bone marrow cells. Fluorescence in situ hybridization using dual fluorescence probe on interphase cells is a new technique for analysis the
chromosomal abnormalities.
@ES Methods :
@EN FISH was performed using the double mixture of DNA probe (Oncor, Gaithersburg, USA) on interphase cells of three patients with chronic myelocytic leukemia and two patients with acute promyelocytic leukemia. Fluorescent signals were detected
by
the
triple band pass filter(Applied Imaging UK) after hybridization and amplification).
@ES Results :
@EN Fused signals were detected more than 90% of interphase cells of all CML patients. In APL patients, the PML-RAR fused signals were identified more than 75% of interphase cells. But the control bone marrow cells showed less than 3% fused
signals.
@ES Conclusions :
@EN FISH using dual fluorescence probe was very simple and useful for detection of fused gene on interphase nuclei. But the probes were too expensive for use in routine laboratory (Korean J Clin Pathol 1995;15(1) : 146~51).
